Integrated modeling of genotype-phenotype relationship in CHO cells

Despite successful efforts in bioprocess optimization, culture media optimization, and targeted metabolic engineering, several protein-based therapeutics remain difficult to express at commercially-enabling productivities and/or appropriate product quality. Quality by design (QbD) efforts involving modeling studies have principally focused on cell metabolism, since it provides the necessary components for protein production and influences various CQAs. Cellular metabolism, however, is just one key aspect of several processes that impact product quality in CHO cells. It is therefore essential to gain a more comprehensive understanding of CHO metabolism and the associated pathways to optimize cell culture bioprocesses and guide cell engineering. In order to facilitate this unmet requirement, my postdoctoral project focus on the development of a systems biology platform to study the processes underlying mammalian cell growth and protein synthesis.
